Choosing a glass tube of 6cm of which the bottom diameter is 3mm, with a small cotton ball inside, making the bottom of the cotton ball and bottom of glass paralleled, and then dropping into the glass tube with 90% of the solution of carbonic acid until the small cotton ball soaked. Then lower end of the glass tube placed at the left side of the mouth of the buccal mucosa, burning for 30s, see the mucosa appeared in the area of about white damage of 3mm. After 24 hours, the rat left cheek test area formed a diameter of 30 ~ 35mm round or oval ulcer, about 0.5mm deep and the surface covered with off-white pseudomembrane. Observing the flushing, swelling, and pain of oral mucosa for several days after dosed separetly, measuring the temperature of mouth and ulcer surface diameter with a vernier caliper and then analyze statistically.
